日粮中不同槲皮素水平对凡纳滨对虾抗副溶血弧菌性能的影响

摘要:
本研究旨在探讨不同槲皮素水平对凡纳滨对虾(Litopenaeus vannamei)抗副溶血弧菌(Vibrio parahaemolyticus,VP)性能的影响。选取凡纳滨对虾(3.65 ± 0.93 g),每100 g商品饲料中分别添加0 g(对照组)、0.1 g、1 g和2 g槲皮素。结果表明:随着饲料中槲皮素水平的升高,对虾肝胰腺超氧化物歧化酶(SOD)活性显著升高(P<0.05),其中1 g和2 g槲皮素组SOD活性显著高于0 g和0.1 g槲皮素组(P<0.05),但1 g和2 g槲皮素组间SOD活性无显著差异(P>0.05)。1 g槲皮素组溶菌酶(LZM)、谷胱甘肽过氧化物酶(GSH-Px)和碱性磷酸酶(AKP)活性显著高于0g、0.1 g和2 g槲皮素组(P<0.05);VP攻毒后,随着饲料中槲皮素水平的升高,对虾肝胰腺LZM活性显著升高(P<0.05),其中1 g和2 g槲皮素组显著高于0 g、0.1 g槲皮素组(P<0.05),但1 g和2 g槲皮素组间无显著差异(P>0.05)。1 g槲皮素组SOD、GSH-Px和AKP活性显著高于0.1 g和2 g槲皮素组(P<0.05);1 g槲皮素组对虾存活率显著高于0.1 g和2 g槲皮素组(P<0.05)。1 g槲皮素组对虾肝胰腺和肠组织免疫基因Toll样受体(TLR)、抗脂多糖因子(ALF)、溶菌酶(lysozyme)、组织蛋白酶B(cathepsin B)、甲壳素(crustin)和转录因子激活蛋白-1(AP-1)的相对表达量显著高于0.1 g和2 g槲皮素组(P<0.05)。综上所述,饲料中添加1 g槲皮素显著增强凡纳滨对虾抗VP性能。槲皮素有望用作虾的功能性饲料添加剂,降低VP感染对虾导致的死亡率。

Effect of different levels of quercetin in diets on anti-Vibrio parahaemolyticus performance in the Litopenaeus vannamei

Abstract:
The aim of this study was to investigate the effect of different quercetin levels on the resistance to Vibrio parahaemolyticus (VP) in Litopenaeus vannamei. Litopenaeus vannamei (3.65 ± 0.93 g) were selected and supplemented with 0 g (control), 0.1 g, 1 g, and 2 g of quercetin per 100 g of commercial feed. The results showed that the superoxide dismutase (SOD) activity of shrimp hepatopancreas increased significantly (P<0.05) with the increase of quercetin level in the feed. The SOD activity of the 1 g and 2 g quercetin groups was significantly higher than that of the 0 g and 0.1 g quercetin groups (P<0.05), but there was no significant difference in SOD activity between the 1 g and 2 g quercetin groups (P>0.05). Lysozyme (LZM), glutathione peroxidase (GSH-Px), and alkaline phosphatase (AKP) activities of the 1 g quercetin group were significantly higher than those of the 0 g, 0.1 g, and 2 g quercetin groups (P<0.05). After VP attack, with the increase of quercetin level in the feed, shrimp hepatopancreatic LZM activity increased significantly (P<0.05). The 1 g and 2 g quercetin groups were significantly higher than the 0 g and 0.1 g quercetin groups (P<0.05), but there was no significant difference between the 1 g and 2 g quercetin groups (P>0.05). SOD, GSH-Px, and AKP activities were significantly higher in the 1 g quercetin group than in the 0.1 g and 2 g quercetin groups (P<0.05). The survival rate of shrimp in the 1 g quercetin group was significantly higher than that in the 0.1 g and 2 g quercetin groups (P<0.05). The relative expression of Toll-like receptor (TLR), anti-lipopolysaccharide factor (ALF), lysozyme, cathepsin B, crustin, and activator of transcription factor-1 (AP-1) in the hepatopancreatic and intestinal tissues of shrimp in the 1 g quercetin group was significantly higher than that in the 0.1 g and 2 g quercetin groups (P<0.05). In conclusion, the addition of 1 g of quercetin to the feed significantly enhanced the anti-VP performance of shrimp Litopenaeus vannamei. Quercetin has the potential to be used as a functional feed additive for shrimp to reduce the mortality caused by VP infection.
