本研究旨在从1株海南霸王岭热带雨林土壤来源的蜡样芽孢杆菌Bacillus cereus BWL-001中挖掘抗海洋弧菌的活性代谢产物,采用多种现代色谱分离方法对菌株提取物进行分离纯化,通过核磁共振波谱解析确定单体化合物结构,并结合96孔法进行抗弧菌活性测试。结果显示,从该菌株LB培养基发酵产物中分离鉴定出10个单体化合物,包括4个二酮哌嗪类化合物(1-4)、3个吲哚生物碱衍生物(6-8)以及3个其他类化合物(5,9和10),分别命名为3-异丁基吡咯并哌嗪-2,5-二酮(1)、cyclo-(Pro-lle ) (2)、maculosin (3)、trans-cyclo-(Pro-Phe) (4)、anthranilic acid (5)、indole-3-meth-ylethanoate (6)、indole-3-acetic acid (7), nb-acetyltryptamine (8)、thymine (9)、acetamide (10)。其中,化合物6-8对副溶血弧菌(Vibrio parahaemolyticus)有弱 抑制活性,其最小抑菌浓度(Minimal Inhibit Concentration,MIC)值均为12.5 μg/mL。本研究丰富了海南霸王岭热带雨林土壤来源的蜡样芽孢杆菌代谢产物多样性,并为抗弧菌活性先导化合物的发现提供理论基础。 |

Study on metabolites and anti-vibrio activity of Bacillus cereus BWL-001 |

The aim of this study was to extract anti-vibrio metabolites from a strain of Bacillus cereus BWL-001 from the soil of Bawangling tropical rain forest in Hainan Province. A variety of modern chromatographic separation methods were used to isolate and purify the extracts of the strain. The structure of the monomeric compounds was determined by NMR spectroscopy. The anti-vibrio activity was tested by 96-well method. The results showed that 10 monomer compounds were isolated and identified from LB culture medium of the strain, including four diketopiperazines (1-4), three indole alkaloid derivatives (6-8), and three other compounds (5, 9 and 10). Named 3-isobutylpyrrolopiperazine-2, 5-dione (1), cyclo-(Pro-lle) (2), maculosin (3), trans-cyclo-(Pro-Phe) (4), anthranilic acid (5), indole-3-meth-ylethanoate (6), indole-3-acetic acid (7), nb-acetyltryptamine (8), thymine (9), acetamide (10). Compounds 6-8 showed weak inhibitory activity against Vibrio parahaemolyticus with MIC values of 12.5 μg/mL. This study enriched the diversity of metabolites of Bacillus cereus derived from soil in Bawangling tropical rain forest in Hainan Island, and provided a theoretical basis for the discovery of lead compounds for anti-vibrio activity. |
