青钱柳叶绿体基因组特征分析

青钱柳(Cyclocarya?paliurus)为我国特有珍贵树种,具有重要的药用、材用和观赏价值,但目前对青钱柳植物研究相对较少。本研究采用Illumina二代测序技术对青钱柳叶绿体基因组进行测序,并对其序列特征、密码子偏好性、重复序列、简单重复序列、系统发育进行分析,结果表明:青钱柳叶绿体基因组大小为161,291 bp,具有典型的环状四分体结构,包含90,367 bp的大单拷贝区,18,590 bp的小单拷贝区和26,167 bp的反向重复区,总GC含量为36.0%;共注释得到113个基因;共鉴定出136个长重复序列,82个简单重复序列(SSR);其密码子偏向使用A/U两种碱基;与化香树属的亲缘关系较近,且支持率为100。研究结果为青钱柳分子标记的开发、物种鉴定、遗传多样性分析和种质资源保护和开发利用提供基础数据。

Abstract:
Cyclocarya paliurus, a precious species endemic to China, has important medicinal, timber and ornamental value. In this study, the chloroplast genome of C. Paliurus was sequenced by Illumina next-generation sequencing technology, and its sequence characteristics, codon preference, sequence repeats, simple sequence repeats and phylogeny were analyzed. The results showed that the chloroplast genome of C. paliurus was 161,291 bp in size, with a typical circular quadripartite structure, including a large single copy region of 90,367 bp, a small single copy region of 18,590 bp and a reverse repeat region of 26,167 bp. The total GC content was 36.0%. The chloroplast genome encoded 113 genes and 136 long repeat sequence, and 82 simple sequence repeat (SSR) were identified. The codon tends to use A/U and the phylogenetic analysis show that C. paliurus had a close relationship with Platycarya?with a strong support (BS =100). This study, enriched the information on the genetic resources of C.paliurus, and provided reference for molecular marker development, species identification, genetic diversity analysis, and germplasm resources protection, development and utilization of C. paliurus.
