织锦巴非蛤育苗期水体菌群结构特征

摘要:
【目的】本文对织锦巴非蛤幼体发育各阶段水体菌群的组成和结构特征进行了研究,为织锦巴非蛤苗种规模化人工繁育提供理论依据。【方法】实验采用Illumina MiSeq高通量技术对幼体在D形幼虫期HD、壳顶期HK、变态前期HB、变态后HA、稚贝期HZ水体菌群进行了测序。【结果】α-多样性指数表明D形幼虫期菌群丰度和菌群多样性均最高,而稚贝期菌群丰度最低,变态前期菌群多样性最低。在门分类水平上,幼体发育所有阶段水样中占优势的细菌门类主要有变形菌门的α-和γ-变形菌纲、拟杆菌门和放线菌门;在科水平上,红杆菌科和黄杆菌科是整个育苗周期的主导类群。微杆菌科相对丰度在变态前期低于1%,在幼体发育后期增长,分别为9.95%和12.82%;冷形菌科则在D形幼虫期不足1%,在HK、HA和HZ阶段的相对丰度较高,在5.98%~7.67%之间。值得注意的是Idiomarinceae科,相对丰度在织锦巴非蛤幼体发育不同阶段有较大差异,在D形幼虫期和壳顶期的相对丰度不足1%,但在变态前期迅速增加到31.90%。【结论】织锦巴非蛤幼体发育不同阶段水体菌群多样性和组成发生了明显变化。

Characterization of aquatic bacterial community of Paphia textile larvae during hatchery period with high-throughput sequencing

Abstract:
【Objective】 In the study, the composition and structural characteristics of the aquatic microbiota of Paphia textile larvae at each developmental stages was investigated, providing a theoretical basis for the large-scale artificial breeding of seedlings.【Methods】The experiment used Illumina MiSeq high-throughput technology to sequence the aquatic microbiota during larvae development at the D-shaped larval stage HD, umbo larvae stage HK, pre-metamorphosis stage HB, post-metamorphosis stage HA, and juvenile stage HZ.【Results】The α-diversity index indicated that the abundance and diversity of microbial communities in the D-shaped larval stage were the highest, while the abundance in the juvenile stage was the lowest, and the diversity in the pre-metamorphosis was the lowest. Bacterial communities were dominated by the Proteobacteria (including α- and γ- Proteobacteria), Bacteroidetes, and Actinobacteriota at phylum level across five larval development stages. At the family level, the Rhodobacteriaceae and Flavobacteriaceae were the dominant groups throughout the larvae development stages. The relative abundance of Microbacteriaceae was less than 1% in the pre-metamorphosis stage, and increases in the late stage of juvenile development, reaching 9.95% and 12.82%, respectively. The Cryomorphaceae family had a relative abundance of less than 1% in the D-shaped larval stage, and was higher in the HK, HA, and HZ stages, ranging from 5.98% to 7.67%. It is worth noting that the Idiomalinceae family had significant differences in relative abundance at different stages of juvenile development of the Paphia textile. The relative abundance was less than 1% during the D-shaped larval stage and umbo larvae stage, but rapidly increased to 31.90% during the pre-metamorphosis. 【Conclusion】There were a marked differences in bacterial community diversity and structure in rearing water during larvae development stages of the Paphia textile.
