傅里叶红外光谱法测定罗汉果中罗汉果总甙含量

摘要:
利用傅里叶变换红外光谱(FTIR)技术建立一种测定罗汉果中罗汉果总甙含量的新方法,通过最小二乘法拟合罗汉果总甙标准品红外光谱1059cm-1特征峰强度的标准曲线,得到回归方程,根据回归方程计算罗汉果样品甲醇超声波提取出物中的总甙含量。将计算结果分别进行精密度、重复性和稳定性检验,平行测定6次,罗汉果总甙的峰高相对标准偏差(RSD)分别为5.41%、5.47%和5.47%;FTIR法测定不同罗汉果样品的总甙含量与紫外分光光度计法的测定结果差异不显著(P>0.05)。本方法具有可行性与可靠性,能够满足实验室的准确度要求,可以作为罗汉果总甙定量测定的方法。

Abstract:
Fourier transform infrared spectroscopy (FT-IR) was utilized as a new approach to investigate the mogrosides contents of Siraitia grosvenorii.A serial intensity value of 1059 cm-1 peak, which was intended to be the characteristic peak of mogrosides within its IR fingerprint spectra, was obtained from the IR spectra of different contents of mogrosides standards and used to draw standard curve as a function of mogrosides contents, in which the curve was fitted with least square fitting (LSF) for establishing linear regression equation.The concentration of mogrosides extracted from fructus momordicae sample with methanol combining ultrasonic method was determined by using the regression equation.The relative standard deviation (RSD) was calculated from 6 replicate determinations, of which the precision, repeatability and stability were 5.41%, 5.47% and 5.47%, respectively.The mogrosides contents measured from different Siraitia grosvenorii samples by FT-IR were not significant differences compared with those determined by UV spectrophotometry (P> 0.05), suggesting that FT-IR is a feasible method for quantificational detection of mogrosides content in Siraitia grosvenorii.
