在3000L发酵罐中,利用分批补料培养技术高密度培养含分泌型重组质粒p ETGAU-10的大肠杆菌TG1,生产α-乙酰乳酸脱羧酶。通过控制发酵条件,发酵液的细胞密度(OD600)可达30.5,且胞外α-乙酰乳酸脱羧酶活力最高达890U·ml-1,达到较为理想的工业化生产的要求。 |

Abstract: |
Escherichia coli TG1,which contained secreted recombinant plasmid pETGAU-10,was cultured to a high density to produce α-ALDC by the fed-batch culture in 3000 L fermentor. In control of the fermentative conditions,the cell density can reach as high as 30.5 of OD600 and the highest extracellular activity of α-ALDC reached 890 U·ml-1,which meet the demand for the ideal industrial production. |
