以薯蓣皂苷元为对照品,采用三氯甲烷-乙酸乙酯(10:1)为展开剂,对绵萆薢药材进行薄层色谱鉴别;采用Hypersil ODS2 C18柱(5μm,4.6mm×250mm),乙腈-水(90:10)为流动相,203nm为检测波长,测定绵萆薢药材中薯蓣皂苷元的含量。绵萆薢药材薄层色谱鉴别中,薯蓣皂苷元与其他成分分离良好,能较好的鉴别出薯蓣皂苷元;HPLC定量分析中,薯蓣皂苷元进样量2.16~19.44μg范围内呈良好的线性关系,平均回收率为100.92%,RSD为1.30%。本方法简便,结果准确,重现性好,可以用于绵萆薢药材的质量控制。 |

Abstract: |
Diascorea septemloba Thunb were identified by TLC respectively;using diosgenin as reference substances, and (10:1)as mobile phases.Diosgenin contents were determined by HPLC using the ODS reversed-phase column of Hypersil C18 (5μm, 4.6mm×250mm), the mobile phase was a combination of acetonitrile and water (90:10), and the eluate was monitored with UV absorption at 203nm.Diosgenin was well-separated by TLC.The diosgenin contents in D.septenloba Thunb by HPLC, in the linear range of 2.16 to 19.44μg and the recoveries of 100.92% (RSD=1.30%).The established methods are simple, feasible and reproducible.It is also proposed to be used for the quality control of the D.septemloba Thunb. |
