超声波协同酶法提取菝葜中总黄酮工艺参数优化

摘要:
[目的]为开发菝葜(Smilax china)资源提供理论和实践依据。[方法]采用超声波辅助酶法提取菝葜中总黄酮,通过单因素试验考察酶用量、pH值、酶解时间和酶解温度对菝葜中黄酮得率的影响。在此基础上,再利用响应面法优化提取条件,研究pH值、酶解时间和酶解温度3个自变量之间的交互作用对黄酮得率的影响。[结果]最佳提取工艺条件为pH值5.10,酶解时间48 min,酶解温度为53℃,在此条件下总黄酮得率达到2.289%。[结论]该优化工艺合理可行,可用于菝葜中总黄酮的提取。

Optimizing of Ultrasonic Assisted Enzymatic Extraction of Total Flavonoids from Smilax china

Abstract:
[Objective] To provide theoretical and practical foundations for the exploitation of Smilax china resources, the total flavonoids in Smilax china were extracted by ultrasonic assisted enzyme method.[Methods] The effects of enzyme dosage, pH value, hydrolysis time and hydrolysis temperature on the yield of flavonoids in Smilax china were investigated in single factor experiments.On the base of this, the optimum conditions for the extraction of total flavonoids from Smilax china were determined by using response surface methodology. And the effects of interaction between pH, hydrolysis time and enzymatic hydrolysis on the yield of flavonoids were studied.[Results] The optimum conditions were as follows:pH was 5.10, hydrolysis time was 48 min, and hydrolysis temperature was 53℃.Under these conditions, the total yield of flavonoids reached 2.289%.[Conclusion] The optimal technology is reasonable and feasible and can be used for the extraction of total flavonoids from Smilax china.
